Their characteristics indicate that the small (95 kD) Akap200 protein is a member of the myristoylated alanine-rich C kinase substrate protein family, whereas the large (200 kD) Akap200 protein is a chimeric A kinase anchor protein-myristoylated alanine-rich C kinase substrate protein. Both large and small Akap200 proteins are myristoylated, but are not palmitoylated. N-terminal myristoylation correlates with the intracellular targeting of the 200 kD Akap200 protein: epitope-tagged 200 kD Akap200 protein is targeted to the surface of S2 cells, while epitope-tagged Akap200 protein missing the first 7 amino acids is distributed throughout the cytoplasm. The PSD (phosphorylation site domain) at amino acids 119-148 of both Akap200 proteins is able to bind with F-actin and calmodulin; this domain can be phosphorylated in vitro by protein kinase C, and phosphorylation interferes with the interaction of the PSD with F-actin and calmodulin. The Pka-R2 tethering domain of the 200 kD Akap200 protein is located at amino acid residues 511-531.
